肉毒桿菌神經毒素（BoNT）是人類已知的毒性大的物質之一。已經鑒定了七種免疫學上不同的神經毒素血清型，命名為A至G型（BoNT / A，BoNT / B，BoNT / C，BoNT / D，BoNT / E，BoNT / F，BoNT / G）。這些肉毒桿菌神經毒素天然產生與一種或多種無毒神經毒素相關蛋白（NAP）復合。NAP對于攝入毒性很重要，因為它們保護神經毒素免受蛋白酶，酸性，熱的影響，并且它們也可能在粘膜層的易位中發(fā)揮作用。
合成為單個150kDa多肽鏈，隨后通過切割激活復合物的實際神經毒素部分以產生兩條鏈，重鏈和輕鏈，其通過單個二硫鍵連接。對于每種毒素，50kDa輕鏈是鋅??依賴性蛋白酶，其切割神經傳遞期間突觸小泡膜融合所必需的單個靶蛋白。A，C和E型神經毒素特異性結合并選擇性地切割突觸體相關蛋白SNAP-25，而B，D，F(xiàn)和G型切割突觸小泡蛋白-2，也稱為VAMP-2。靶蛋白的切割抑制神經元之間的神經遞質釋放，這導致肌肉麻痹。
肉毒桿菌神經毒素毒性
每種毒素很容易分為三個區(qū)域，反映出對毒性很重要的三個功能; 特異性底物的結合，易位和切割。重鏈負責識別突觸前膜上的特定神經節(jié)苷脂和特定蛋白質受體。重鏈的易位結構域部分介導輕鏈穿過內體膜，后靶蛋白被酶促結構域切割。
肉毒桿菌神經毒素在研究中的應用
肉毒桿菌神經毒素是研究中有價值的研究工具，旨在闡明囊泡運輸和毒性所涉及的機制，以及了解突觸傳遞的潛在事件。除了使用150kD全毒素的研究之外，重組無毒重鏈可用于評估毒素的結合特性，并且重組無毒輕鏈可用于篩選抑制蛋白酶結構域的酶活性。
